Q Exactive™ HF-X Mass Spectrometer
Description
The Q Exactive™ HF-X is a high-resolution hybrid benchtop mass spectrometer. It integrates a linear quadrupole for highly precise precursor ion selection, a Higher-Energy Collisional Dissociation (HCD) cell for high-energy fragmentation, and an Orbitrap Fourier transform mass analyzer for high-resolution detection of peptides and/or their fragments.
Thanks to the flexibility of its scan modes, this instrument supports both data-dependent acquisition (DDA) for protein identification and targeted analyses (at both MS and MS/MS levels).
The Q Exactive™ HF-X is coupled to a nano-Ultra High Performance LC system (NanoLC RSLC U3000, Thermo Fisher Scientific), specifically designed for nanoscale applications.
Commissioned in September 2018.
Applications
- Protein identification from simple or complex mixtures (bottom-up proteomics)
- Relative quantification of low-abundance proteins in complex samples (label-free, SILAC, TMT)
- Targeted quantification (PRM)
Requirements / Constraints
This instrument is not open access
Funding
The Q Exactive™ HF-X (€500k) was funded through the Biomarkers and Diagnostics Initiative in Languedoc-Roussillon (IBDLR), led by the Biosé – François Rabelais cluster and selected within the State-Region Contract Plan (CPER) 2015–2020.
The associated nano-UHPLC system was funded by the UAR BioCampus Montpellier (small equipment program 2017, €22.5k) and internal funds from the FPP platform (€25k).
