Quantitative Proteomics
Description / Workflow
Quantification is based on
- Label-free approaches: Samples are analyzed independently by LC-MS. This method requires multiple replicates and relies on the stability and reproducibility of the nanoLC-MS system.
- Isotopic labeling approaches: A mass tag containing heavy isotopes is introduced, either at the peptide level via isobaric labeling (TMT/iTRAQ) or at the protein level via metabolic labeling (SILAC). Samples can then be pooled and analyzed simultaneously.

Applications
- Basic and/or clinical research
- Study of proteome variations
Requirements / Constraints
- Selection of experimental design (label-free, SILAC, TMT, etc.) guided by sample type (cell cultures, tissues, etc.), complexity, and available material
- Sufficient number of experiments for statistical pre-processing
- High-resolution mass spectrometer
Technologies
- Label-free
- Chemical labeling (TMT/iTRAQ)
- Metabolic labeling (SILAC)
