Quantitative Proteomics

Description / Workflow

Quantification is based on

  • Label-free approaches: Samples are analyzed independently by LC-MS. This method requires multiple replicates and relies on the stability and reproducibility of the nanoLC-MS system.
  • Isotopic labeling approaches: A mass tag containing heavy isotopes is introduced, either at the peptide level via isobaric labeling (TMT/iTRAQ) or at the protein level via metabolic labeling (SILAC). Samples can then be pooled and analyzed simultaneously.

Applications

  • Basic and/or clinical research
  • Study of proteome variations

Requirements / Constraints

  • Selection of experimental design (label-free, SILAC, TMT, etc.) guided by sample type (cell cultures, tissues, etc.), complexity, and available material
  • Sufficient number of experiments for statistical pre-processing
  • High-resolution mass spectrometer

Technologies

  • Label-free
